On November 22, within the framework of the 2024 Technology Exchange, the Center for Science and Technology Information and Statistics (CESTI), in collaboration with the University of Medicine and Pharmacy in Ho Chi Minh City, organized a technology cooperation event themed “Production of Herbal Capsules for Insomnia Treatment from Mimosa pudica, Erythrina variegata, Magnolia officinalis (Southern variety), and Glycyrrhiza uralensis (Southern licorice)”, with the participation of representatives from T&T Pharmaceutical Machinery Co., Ltd. as the technology consulting partner.
Research on formulation and evaluation of the sedative effect of Production of Herbal Capsules for Insomnia Treatment using Mimosa pudica, Erythrina variegata, Magnolia officinalis (Southern variety), and Glycyrrhiza uralensis (Southern licorice).
Vietnam possesses a vast resource of medicinal plants that are widely used as raw materials for the production of health-care and wellness products. Among them, herbs such as Mimosa pudica (Xấu hổ), Erythrina variegata (Vông nem), Magnolia officinalis – Southern variety (Hậu phác nam), and Southern licorice (Cam thảo nam) are natural medicinal sources known for their safe, mild, and effective sedative and insomnia-relieving properties.
These herbs have been applied in the study titled: “Production of hard capsules with sedative effects for insomnia treatment from Mimosa pudica, Erythrina variegata, Magnolia officinalis (Southern variety), and Southern licorice”, opening new directions for product development and offering a more effective solution for managing insomnia symptoms.
This research is the result of a scientific project led by the University of Medicine and Pharmacy in Ho Chi Minh City, which was officially accepted and approved by the Ho Chi Minh City Department of Science and Technology in 2024.
At the event, Dr. Hứa Hoàng Oanh (representing the research team from the University of Medicine and Pharmacy in Ho Chi Minh City) shared that the production process for the herbal-based insomnia-treatment capsules was developed from a traditional folk remedy that had been effectively applied in practice by Dr. Võ Đình Hưng. Through this study, the research team aims to further develop and enhance the efficacy of the remedy by optimizing the extraction process and conducting quality control to ensure maximum safety and therapeutic effectiveness.
Research Content: Formulating Herbal Capsules for Insomnia Treatment
Dr. Hứa Hoàng Oanh explained that insomnia can arise from various causes, and several studies have shown a strong correlation between symptoms of depression, anxiety, stress, and insomnia. Additionally, research indicates that approximately 19–74% of insomnia-related symptoms are associated with psychological factors. Besides psychological therapy, the use of sedative medication plays an important role in treating insomnia.
In the domestic market, many sedative and anxiolytic products originate from synthetic pharmaceuticals. Although these medications are effective, their duration of use is limited, and they often cause unwanted side effects, including tolerance and dependence. Furthermore, most herbal-based products rely on imported ingredients, making quality control difficult and increasing production costs.
The research team led by Dr. Hứa Hoàng Oanh aims to standardize the herbal extracts used in the traditional formula composed of Mimosa pudica (Xấu hổ), Erythrina variegata (Vông nem), Magnolia officinalis – Southern variety (Hậu phác nam), and Glycyrrhiza uralensis (Southern licorice) (Cam thảo nam). This is intended to ensure consistent quality during extract production, contributing to product safety and therapeutic efficacy. At the same time, the team sought to prepare hard capsules derived from the decocted herbal formula with sedative and insomnia-relief properties.
According to Dr. Hứa, several studies have demonstrated the therapeutic effects of these herbs: Mimosa pudica helps reduce anxiety and depression and enhances memory; Southern licorice has sedative and anxiolytic properties; and Erythrina variegata exhibits anxiolytic and antidepressant effects.
Chemical compounds found in the herbal materials, such as mimosine (present in Mimosa pudica) and coixol (a biologically active compound in Southern licorice), were used as marker substances for quality control of the formula.
The herbal extract was preserved using potassium sorbate, added within permitted limits. Developing the formulation and production process for herbal insomnia-relief capsules addresses the needs of the Vietnamese pharmaceutical industry by gradually substituting expensive imported proprietary medicines and promoting domestic pharmaceutical development.
In this study, the research team conducted five main tasks:

Testing and evaluating raw herbal materials.

Extracting herbal actives and establishing quality standards for the extracts.

Investigating effective sedative and anxiolytic doses of the extracts.

Formulating the capsule product.

Assessing safety and evaluating sedative, anxiolytic, and antidepressant effects of the product on white mice subjected to isolation-induced stress.
Research Results: Formulation of Herbal Sedative Capsules for Insomnia Treatment
The results showed that the aqueous extract (freeze-dried to 0 percent moisture) exhibited no oral toxicity at a maximum dose of 18.16 g/kg body weight in experimental mice.
Male and female white mice were administered the hard-capsule sedative formulation at doses equivalent to 1 capsule/kg to 2 capsules/kg, once daily for 60 consecutive days. The mice developed normally, showing no abnormalities in behavior, general condition, liver and kidney function, or hematological parameters. The product demonstrated sedative and antidepressant effects, was safe, and showed no signs of toxicity.
Formula and Production Process for Herbal Sedative Capsules
After evaluating suitable excipients and optimal ratios, the team successfully developed a hard-capsule formulation. Three laboratory-scale batches of hard capsules were produced, each containing 1,000 capsules.
A pilot-scale manufacturing process was then established and tested on three research batches of 15,000 capsules each. The team also developed internal quality standards for the hard capsules in accordance with the guidelines of the Vietnamese Pharmacopoeia V, and conducted stability studies under real-time storage conditions as well as accelerated aging conditions.

Dr. Hứa Hoàng Oanh stated that the outcomes of this project will serve as valuable scientific and practical reference materials for research and manufacturing units. The findings also provide a foundation for upgrading the standards and quality of domestic herbal extracts and herbal-derived products. Furthermore, the successful formulation of capsules and the evaluation of their sedative effects from the herbal insomnia-treatment decoction containing Mimosa pudica, Erythrina variegata, Magnolia officinalis (Southern variety), and Glycyrrhiza uralensis (Southern licorice) in Vietnam can help replace imported products, reduce production costs, enhance treatment effectiveness, and offer greater convenience for users. |

At the event, interested organizations shared valuable insights regarding opportunities for cooperation and the practical implementation of technology transfer in producing herbal insomnia-relief capsules. The discussions focused on key issues such as potential side effects, drug-dependence risks, product registration procedures, clinical trials, and production costs.
